The Oris Pro Cell Invasion Assay enables the use of automated liquid handling equipment for cell seeding and allows unlimited access to cells throughout the experiment. The Oris Pro Cell Invasion Assay uses a non-toxic biocompatible gel, BCG (see Figure 1: Well Bottom with BCG), to form a centrally located and temporary cell-free zone on culture surfaces. Cells are seeded into the 96-well plate and attach in a monolayer around the BCG. The BCG dissolves to reveal a Detection Zone and a Collagen I Overlay is added to create a 3-D ECM environment for cell invasion (see Figure 2: Assay Schematic). Cells can be treated with one or multiple stains or labels. The cells in the Detection Zone can be quickly captured and easily quantified in real-time using microscopes and High Content Screening (HCS)/High Content Imaging (HCI) instruments. Cell invasion above the plate surface can be assessed by performing a Z-stack analysis. Staining live cells at the beginning of the experiment permits real-time tracking of cell movement that can be captured at any time point. Both endpoint and kinetic data can be generated.

Figure 3: Oris Pro Collagen I Cell Invasion Analysis: HT-1080 cells were seeded onto an Oris Pro Collagen I plate. Following the addition of a 3.0mg/mL Collagen I gel overlay, cell invasion was permitted for 72 hours in the presence of 10% FBS. Cells were fixed and stained with TRITC-phalloidin and DAPI. Images were captured using the BD Pathway 855 Bioimaging System and the number of invading cells in the Detection Zone was quantified at defined Z intervals. Based on a Z-stack analysis of 8 wells, 416 cells +/- 38 invaded 60-150 um into the Collagen I Overlay in the Detection Zone. Images of pre-invasion (A) and invasion at 72 hr (B-E) are shown (phalloidin pseudo-colored yellow), in addition to images at Z-intervals of 60 um (C) and 150 um (D) into the Collagen I gel. A color-coded composite of Z-stacks at intervals of 60, 90, 120, and 150 um obtained from a single well is shown (E).
